ANA Screen ELISA kit Background Information:
Anti-nuclear antibodies (ANAs), also known as anti-nuclear factor or ANF are autoantibodies in blood and produced by immune system which also attack the body's own tissues by works against one or more protein in the body. Body normally produces antibodies to attack and destroy the invaders or foreign organisms such as virus and bacteria. ANAs are abnormal antibodies or autoantibodies with the capability of binding to various parts of the nucleus of the cells and working against the body. The tendency of the immune system to work against its own body is referred to as autoimmunity. Thus, ANAs may be indicative of the presence of autoimmune disease when detected. Autoimmune diseases are disorder of the immune system characterized by the abnormal production of auto-antibodies directed against the tissues of the body. Autoimmune diseases characteristically feature inflammation of various tissues of the body. ANAs are found in patients with a number of different autoimmune diseases, such as systemic Lupus erythematosus, drug induced lupus, Sjogren's syndrome, Rheumatoid arthritis, Scleroderma, Vitiligo, Hashimoto thyroiditis, Juvenile diabetes mellitus, Addison disease, pernicious anemia, glomerulonephritis, and pulmonary fibrosis. Moreover, ANAs can be found in patients with conditions that are not considered typical autoimmune diseases, such as Cancer and chronic infections. The ANA elisa test detects the autoantibodies present in serum. Autoantibody screening is useful in the diagnosis of autoimmune disorders and monitoring levels helps to predict the progression of diseases or assessing the efficacy of a treatment.

ANA Screen ELISA Test kit Principles
ANA Screen ELISA test system is designed to detect IgG class antibodies to a variety of common nuclear antigens in human sera. Wells of plastic microwell strips are sensitized by passive absorption with antigen. The test procedure involves three incubation steps:
1. Test sera are incubated in antigen coated microwells. Any antigen specific antibody in the sample will bind to the immobilized antigen. The plate is washed to remove unbound antibody and other serum components.
2. Peroxidase Conjugated goat anti-human IgG (gama chain specific) is added to the wells and the plate is incubated. The Conjugate will react with the antibody immobilized on the solid phase in step 1. The wells are washed to remove unreacted Conjugate.
3. The microwells containing immobilized peroxidase Conjugate are incubated with peroxidase
Substrate Solution. Hydrolysis of the Substrate by peroxidase produces a color change. After a period of time the reaction is stopped and the color intensity of the solution is measured photometrically. The color intensity of the solution depends upon the antibody concentration in the original test sample. For further details please refer to complete instructions manual supplied with the product.

An OD ratio > 1.10 is interpreted as positive for IgG ANA
An OD ratio < 0.90 is interpreted as negative for IgG ANA


ANA Screen ELISA Test kit Precautions
The human serum controls are POTENTIALLY BIOHAZARDOUS MATERIALS. Source materials from which these products were derived were found negative for HIV-1 antigen, HBsAg & for antibodies against HCV and HIV by approved test methods. However, since no test method can offer complete assurance that infectious agents are absent; these products should be handled at the Biosafety Level 2 as recommended.
