Material Provided with Chagas ELISA kit:
1. Test Strips Microwells: 96 test wells containing Trypanosoma cruzi antigens
2. Enzyme Conjugate: anti-human Ig Peroxidase-HRP
3. Positive Control: diluted Trypanosoma cruzi-positive sera in buffer
4. Negative Control: diluted Trypanosoma cruzi-negative sera in buffer
5. TMB Substrate Solution: TMB
6. Wash Concentrate 20X
7. Dilution Buffer
8. Stop Solution: 0.73 M phosphoric acid

Materials Required, Not Provided:
1. Precision pipettes
2. Distilled or deionized water

Chagas ELISA kit Background Information:
Trypanosoma cruzi is a protozoan parasite, which is the causative agent of Chagas disease. This disease ranges from southern United States to Northern Argentina and Chile. The disease is transmitted to humans through the bite wound caused by reduviid bugs, blood transfusions, and in newborns, infection in utero. In acute infections, there may be few or no symptoms of the disease. In chronic infections, there may be inflammatory cardiomyopathy, or severe dilation of the esophagus or colon known as megadisease.

Chagas ELISA Test Principle:
The principle of the Chagas IgG ELISA test involves a two-step incubation procedure. The antibodies in the patient serum bind to the antigens in the test well in the course of the first incubation stage. After this, the enzyme complex binds to the antigen-antibody complex, which occurs in the second incubation step. The unbound enzymes are then removed through repeated washings. In the presence of this enzyme complex and peroxide, a substrate is added, and as a result of this combination, the substrate color will turn blue. The reaction is ended with the stop solution, and what appears is a yellow color instead of the blue. For additional details please refer to the instructions for use.

Notable features of the Parasitology ELISA Kits:
User-friendly directions and explanation of test procedures
Simple and safe reagent preparation
Clear instructions on specimen collection
Comprehensive package of required materials
Explicit quality control and storage guidelines
Reliable and easy-to-read test results

Product inserts for most Parasitology ELISA kits follow a similar method. See the Chagas ELISA Kit product insert for details on preparation, procedures, quality control, and test result interpretation.
