How do doctors find hepatitis C antibodies in the blood? To determine if hepatitis C antibodies are in the blood, doctors use a screening test called ELISA, which stands for Enzyme-Linked Immunosorbent Assay.  There are many different ELISA tests, so doctors must use one specifically for hepatitis C.  This is where the HCV ELISA Kit comes in.  The HCV ELISA Kit will search the blood sample for the hepatitis C antibody.  This HCV ELISA Kit is very sensitive and picks up approximately 95% of people infected with hepatitis C antibodies.  If the HCV ELISA Kit test finds any hepatitis C antibodies, that means an HCV infection is present.  The HCV ELISA Kit is tempered by its sensitivity.  For example, if the HCV ELISA Kit tests negative, patients are most likely free of hepatitis C infection.  However, if the HCV ELISA Kit test is positive, there is a small possibility that the result could be incorrect, resulting in a “false-positive.”  False-positives on this HCV ELISA Kit usually indicate more testing is required.

Clinical studies show that a significant amount of HCV infected individuals develop antibodies to NS5 non-structural protein of the virus.  For this, the third generation HCV ELISA Kit tests include antigens from the NS5 region of the viral genome in addition to NS3 (c200), NS4 (c200) and the Core (c22).  Third generation HCV ELISA KIT tests have improved sensitivity and shorten the time between infection with HCV and the appearance of detectable antibodies (window period) to 60 days.

Hepatitis C virus (HCV) is an envelope, single stranded positive sense RNA (9.5 kb) virus belonging to the family of Flaviviridae. Six major genotypes and series of subtypes of HCV have been identified. Isolated in 1989, HCV is now recognized as the major cause for transfusion associated non-A, non-B hepatitis. The disease is characterized with acute and chronic form although more than 50% of the infected individuals develop severe, life threatening chronic hepatitis with liver cirrhosis and hepatocellular carcinomas. Since the introduction in 1990 of anti-HCV screening of blood donations, the incidence of this hepatitis C infection in transfusion recipients has been significantly reduced.

RESULTS OF HCV ELISA TESTS

 

 

HCV ELISA kit (HCV ab ELISA Kit)

This anti-HCV employs solid phase, indirect ELISA method for detection of antibodies to HCV in two-step incubation procedure. Polystyrene microwell strips are pre-coated with recombinant, highly immunoreactive antigens corresponding to the core and the non-structural regions of HCV (third generation HCV ELISA). During the first incubation step, anti-HCV specific antibodies, if present, will be bound to the solid phase pre-coated HCV antigens. The wells are washed to remove unbound serum proteins, and rabbit anti-human IgG antibodies (anti-IgG) conjugated to the enzyme horseradish peroxidase (HRP-Conjugate) is added. During the second incubation step, these HRP-conjugated antibodies will be bound to any antigen-antibody(IgG) complexes previously formed and the unbound HRP-conjugate is then removed by washing. Chromogen solutions containing Tetramethylbenzidine (TMB) and urea peroxide are added to the wells and in presence of the antigen-antibody-anti-IgG (HRP) immunocomplex, the colorless chromogens are hydrolyzed by the bound HRP conjugate to a blue-colored product. The blue color turns yellow after stopping the reaction with sulfuric acid. The amount of color intensity can be measured and it is proportional to the amount of antibody captured in the wells, and to the amount of antibody in the sample respectively. Wells containing samples negative for anti-HCV remain colorless.

This kit employs indirect ELISA assay for detection of IgM antibodies to HCV in two-step incubation procedure. Polystyrene microwell strips are pre-coated with recombinant, highly immunoreactive antigens corresponding to the core and non-structural regions of HCV. During the first incubation step, HCV-IgM specific antibodies, if present, will be bound to the solid phase pre-coated HCV antigens. The wells are washed to remove unbound materials, and then, rabbit anti-human IgM antibodies (anti-IgM) conjugated to horseradish peroxidase (HRP-Conjugate) are added. The Hepatitis B Test checks for substances in the blood that show whether a hepatitis B infection is active or has occurred in the past.  The Hepatitis ELISA Test Kit (HBsAg ELISA Kit) looks for different signs of infection (markers).  For example, Antigens are markers (HBsAg) made by bacteria or viruses, so the presence of hepatitis B antigens means that the virus is in the body.  Hepatitis B surface antigen (HBsAg) is the earliest sign of an active hepatitis B infection.  This antigen may be present before symptoms of an HBV infection are present.  If this antigen is present for more than 6 months, then there is probably a chronic long-term HBV infection.  This means the Hepatitis B virus can be spread to others through a person’s life.  On the other hand, antibodies (HBsAb) are proteins produced by the body to fight infection.  The presence of Hepatitis B antibodies means that there has been an exposure to the hepatitis B virus, whether from a long past exposure or a current infection.  Genetic material (DNA) of the hepatitis B virus shows that the virus is in the body.  The amount of DNA can help determine how severe the hepatitis infection is and how easily the HBV infection can be spread.  It is important to identify the type of hepatitis virus causing the infection to prevent its spread and choose the proper treatment.

RESULTS OF HBsAg ELISA TESTS

Hepatitis B virus (HBV) is an enveloped, double-stranded DNA virus belonging to the Hepadnaviridae family and is recognized as the major cause of blood transmitted hepatitis together with hepatitis C virus (HCV).   HBV virus infection induces a spectrum of clinical manifestations ranging from mild, inapparent disease to fulminant hepatitis, severe chronic liver diseases, which in some cases can lead to cirrhosis and carcinoma of the liver.
