The Diagnostic Automation Dengue IgM ELISA test is a semi-quantitative enzyme immunoassay for the detection of antibodies to dengue, in samples of human serum or plasma.




Materials Provided with Dengue IgM Elisa test kit:

Dengue IgM Elisa Kit Background Information:
Dengue fever also known as Breakbone fever which is endemic throughout many of the tropical and subtropical regions of the world is caused by any of the four serotypes of dengue virus. The causative agent for dengue fever is a flavivirus and like other types of flavivirus it is transmitted to humans by mosquitoes mainly the Aedes aegypti and A. albopictus type. Dengue fever is the most common viral disease spread to human by mosquitoes. Repeat infection with a second type of dengue virus is thought to cause dengue hemorrhagic fever in about 10 percent of infected people. Antibodies to one type of dengue virus confer immunity to that type, but do not confer immunity to a second dengue type beyond 6-8 weeks. Infection with dengue virus can result in a wide disease spectrum ranging from mild fever to life-threatening dengue hemorrhagic fever and dengue shock syndrome. Common symptoms include fever, chills, skin rash, severe headache, nausea, vomiting, and malaise. In dengue hemorrhagic fever there is a significant depression of platelet levels and increased blood vessel permeability that can lead to shock and death. Dengue fever can only be treated by supportive care, however can be prevented by mosquito control. In primary infections, the first immune response is production of IgM antibodies to the virus, which appear 4-6 days after the onset of symptoms and persist for 2-4 months. IgG antibodies appear approximately 14 days after onset of symptoms and gradually decrease but persist at low levels for life. In secondary infections, IgG antibodies increase to high levels within 2 days of the onset of symptoms, while IgM antibodies may reappear but gradually diminish. These patterns of dengue antibody development permit serological differentiation of primary and secondary infections.



Dengue IgM Elisa Test Principle:
The microwells are coated with purified dengue virus antigen from Vero cell cultured type 1-4 dengue. During the first incubation with the diluted patient sera, any antibodies which are reactive with the antigen will bind to the coated wells. After washing, the Enzyme Conjugate is added. If antibodies have been bound to the wells, the Enzyme Conjugate will then bind to these antibodies. After another series of washes, a chromogen TMB is added. If the Enzyme Conjugate is present, the peroxidase will catalyze a reaction that consumes the peroxide and turns the chromogen from clear to blue. Addition of the Stop Solution ends the reaction and turns the blue color to a bright yellow color. For additional details please refer to the instructions for use.
